Seconding @Bloomer's recommendations of Agents of the Realm and especially Sister Claire, which according to the authors/artists is a comic where literally every character is queer in one way or another as well as just being a fun and wacky fantasy story! Also one of the few media I've seen with an explicitly asexual character which made me very very happy. I'm guessing you're looking for mostly happy/positive stories so I'll put some down here as well! Not all of these are lgbtq+ centered regarding story, but I tried to just list ones where the main characters are lgbt+ and not just random side characters/only hinted at:

Check Please! by Ngozi Ukazu– one of my absolute faves!! About hockey and baking and college with some amazing characters and storytelling and while it has serious bits, is overall just very positive and uplifting. It's a great kind of deconstruction (in a fun way, not a dark and gritty way) of ideas of masculinity in sports, and the main canon couple is amazingly characterized and really adorable too eheh.
Gunnerkrigg Court: famous webcomic for many reasons and pretty long, but later on there are more lgbt+ main characters that are revealed as well who play major roles in the story!
Goth Western: On our very own Tapastic! Really gorgeous art and mysterious story featuring lesbian leads.
Countdown to Countdown: Post apocalyptic/scifi, also really gorgeous art.
Witchy: description according to the author: "Queer Asian witches with hair-powered magic" pretty cool if you ask me
Also well its not the focus but my biblical/historical comic Agapetos features a gay couple as part of the main cast, although since i literally just started last week it's really not as established yet.
uhhh there's a lot more I'm sure!!! I tend to find a lot more lgbtq+ representation in webcomics and other independent media than in mainstream stuff so hyehh.
Other media–I don't watch a whole lot of movies/shows but just a few that I know of!
D.E.B.S. : Teen Spy/action movie featuring a great quirky romance between the heroine and the villainess. It's super cute and while the action and plot is kind of silly it's a load of fun.
Steven Universe: Pretty sure this has been listed at some point since it's become well known for its positive and very overt on-screen portrayals of female/female romantic relationships!! Kind of a hotbed fandom-wise but as a show it's super fun and clever with really good writing.
