If I'm working traditionally, I do a rough thumbnail on a smaller scale to make sure I get the proprtions right. I then blow it up on a copier(working at a office gig gives me perks like using copiers for my art purposes after hours) and lightbox it onto my board; I use non photo blue pencil and do the final [pencil] draft. I then ink it(either with nib & india ink OR microns & brush pen), clean up any "boo-boos", and scan. My finishes are usually clean and mesh with my digital enhancements pretty decent.
However, over the last half year, I've been pushing myself to get better at drawing and inking digitally on Manga Studio. I'll draw(rough layouts) and ink there, then save a version as a JPG and import it to Photoshop to color(yeah I know I can color in MS as well, but I'm not too partial to MS's color in addition to the fact that I'm also still learning MS as program). I like how the digital render looks really solid and crisp- it elevates the level of the colors/lettering as well.
Also when I work traditionally I had to carry around a lot of stuff if I wanted to work on something(bristol, pencil, inks or microns, lightbox). With digital, only thing I carry is my laptop & my Wacom tablet.
