Let's say that I have 43 panels for the introduction of a high school comedy/slice of life.
I read from some sources that I should publish the first three episodes simultaneously so that the reader can get a feel for how the story will go and feel, but I don't know how many panels are supposed to go in each episode!
I could probably do guesswork, but I'm unsure if there's a secret "rule" about this subject.
I don't want the episode to be too long, but I don't want it to be too short, you know?
Should I go off the flow of the story, or should I stop the episode around a certain point?

I have no idea tbh, and I don't think there is a secret rule. I have at least 2-3 panels (on average) on a single page for my Slice of Life series.
I say go with the flow of the story, but do break it up to avoid reader burn out.
Other question is, will it only be page by page updates or multiple page updates

You don't have to split them equally. Try to split them so each episode has some kind of satisfying joke or a moment of emotional drama, or a character reveal or something, so they all feel like a nice chunk of content.

It depends on what style of comic you are writing.

I have seen some with as short as 4 panels (for gag-a-day style slice-of-life) and others that are much longer for ones that have more plot.
