If you are making a comic which is not a gag-a-day and has a "plot", then I suggest focusing more on jokes involving a character interacting with someone or something. I think having a foolish protagonist helps with this. I think the type of thing people find the funniest is that paired with irony.
I am going to recommend avoiding memes and super topical jokes.
Something to watch out for (which people sometimes do) is using "cuteness" as a replacement for a joke or comedy. Like the whole "it's funny because it is a cute baby" or "it's funny because they are cute school girls". You can have cute characters however, them being cute should not be the "set up" for the majority of the jokes. It's not funny, just bland.
