Yes, I actually have more followers on Twitter than I do through Tapastic.
Twitter, much like any other social media site, becomes fun when you find people worth following on the site and start building a feed full of posts that interest you the most. So yes, find your friends on Twitter first, then join communities like #webcomicchat and #comictalk, then think about joining special interest groups (like for me that's is #creaturechat). And promote it some! Tell your readers that you're on twitter and they can follow you there as well.

Share a piece of you every now and then and don't just spam links to your comic. People like following and getting to know other people. So share what you're passionate about, maybe a tip to making comics you've learn, a cute gif/meme, a random fact, just be you, and joining the above mentioned chat groups is a great way to meet the community and mingle/network.
You have 140 characters to blurb something out to the world. It can be challenging when you need more characters to complete a thought, however, just trim it down into the bite size 140 or post the rest of your thoughts in a new tweet. After a while you get used to the character restriction and now I even really appreciate it since I don't spend too long reading long posts on Twitter.
Hashtags are good where appropriate however there's no need to go hashtag crazy especially when you start to form a following. So in the beginning use hashtags like #comics, #webcomics, #webcomicchat, #comictalk. And depending on your post maybe more content related hashtags #fantasy, #humor, #lolz, #scifi, so on so on.
Activity is just your feed wall like on facebook, it's where you can read your friends tweets. Be sure to interact with other people's tweets. Everyone likes getting a like/retweet, however, reading a nice comment means so much more. Do your best to comment on the tweets you like.
Re-posts=Retweets. It's basically sharing someone's tweet like sharing a post on facebook.
